Overview of Gene Research
Trpm2, short for transient receptor potential melastatin 2, is an oxidant-activated non-selective cation channel. It is widely expressed in mammalian tissues. Oxidative stress stimulates intracellular ADP-ribose formation which opens TRPM2 channels, acting as an endogenous redox sensor for mediating oxidative stress/ROS-induced Ca(2+) entry and subsequent Ca(2+)-dependent cellular reactions like endothelial hyperpermeability and apoptosis [2].
In a study on acute myocardial infarction (AMI) in mice, knockout of TRPM2 protected against myocardial injury. Myocardial infarction was induced by ligation of the left anterior descending coronary artery in wild-type and TRPM2 knockout (TRPM2-/-) male mice. Knockout of TRPM2 led to up-regulation of clusterin, suggesting an interaction between the two in AMI [1].
In conclusion, Trpm2 is crucial in mediating oxidative stress-related Ca(2+) entry and cellular responses. The use of TRPM2 knockout mouse models has revealed its role in myocardial injury, specifically in the context of its interaction with clusterin in AMI, providing insights into potential treatment strategies for such cardiac conditions.
