Overview of Gene Research
Ppip5k1, short for diphosphoinositol pentakisphosphate kinase type 1, is an enzyme that generates inositol pyrophosphates (InsPPs). These InsPPs play roles in various cellular processes such as growth factor and insulin signaling [2,4]. Ppip5k1 is involved in pathways related to apoptosis, ligand competition for polyphosphoinositide-binding domains, and cell motility [1,2,3].
Overexpression of Ppip5k1 decreases cell sensitivity to cytotoxic agents like etoposide, cisplatin, and sulindac, and reduces p53 phosphorylation on key residues, suppressing apoptosis [1]. Its catalytic activity is crucial for this anti-apoptotic effect, differentiating it from IP6K2 [1]. In NIH 3T3 cells, stress or receptor activation promotes Ppip5k1 translocation from the cytoplasm to the plasma membrane, modulating ligand competition between diphosphoinositol polyphosphates and PtdIns(3,4,5)P3 for polyphosphoinositide-binding domains [2]. Knockdown of Ppip5k1 in HeLa cells decreases cell motility, suggesting its role in regulating exocyst complex function for cellular polarity and directional migration [3].
