Overview of Gene Research
Ramp2, short for Receptor Activity-Modifying Protein 2, is an accessory protein for certain receptors. It plays a crucial role in various biological processes related to receptor-mediated signaling. It is involved in pathways such as those related to blood sugar homeostasis, vascular and cardiac homeostasis, and angiogenesis. Its function is also linked to the development and function of endothelial cells [1-5].
Vascular endothelial cell-specific Ramp2 knockout mice showed that the AM-Ramp2 system is vital for vascular integrity and homeostasis from prenatal to adult stages [1]. Cardiomyocyte-specific Ramp2 knockout (C-Ramp2-/ -) mice, after transverse aortic constriction, exhibited significant cardiac hypertrophy, decreased ejection fraction, increased fibrosis, reduced ventricular contractility and relaxation, along with enhanced hypertrophy and oxidative stress in cardiomyocytes exposed to isoproterenol. This indicates the AM-Ramp2 system is necessary for early adaptation to cardiovascular stress by regulating cardiac mitochondria [2]. A heterozygous Ramp2 knockout mouse model demonstrated that disruption of the Ramp2/CRLR-cAMP axis could cause primary open-angle glaucoma [3].
In conclusion, Ramp2 is essential for maintaining vascular and cardiac homeostasis. Gene knockout mouse models, especially those targeting Ramp2 in specific cell types, have revealed its role in diseases such as cardiovascular disorders and primary open-angle glaucoma. Understanding Ramp2's function can provide insights into the mechanisms of these diseases and potentially lead to new therapeutic strategies.
