Overview of Gene Research
DERL3, also known as Derlin-3, is a key gene in the endoplasmic reticulum-associated protein degradation pathway. It is involved in the degradation of misfolded lumenal glycoproteins induced by endoplasmic reticulum (ER) stress [7].

DERL3 has been studied in multiple disease contexts. In colorectal cancer, its low expression in cancer tissues is linked to a high rate of lymphatic and distant metastasis. Overexpression of DERL3 in HT29 cells suppresses their proliferative, migratory, and invasive capacities, likely through negatively regulating MYCN level [1].

In gastric cancer, DERL3 acts as a tumor suppressor, with its promoter methylation correlated with tumor clinical features. Genes co-expressed with DERL3 are involved in adaptive immune response, T cell activation, etc. [3].

In nasopharyngeal carcinoma, Epstein-Barr virus latent membrane protein 1 represses DERL3 via DNA methylation, and DERL3 knockdown leads to increased cell proliferation, migration, and invasion [5].

In lung adenocarcinoma, DERL3 is upregulated in carcinoma tissues, and high-expressed patients have a worse response to chemotherapy and immunotherapy. It is positively correlated with B cell activation and unfolded protein response (UPR) [6].

In renal clear cell carcinoma, elevated DERL3 expression indicates a poor prognosis [4].

In rheumatoid arthritis, DERL3 is up-regulated in fibroblast-like synoviocytes of RA patients, and TNF-α-induced DERL3 expression promotes inflammation through activating the NF-κB signaling pathway [2].

In conclusion, DERL3 plays crucial roles in various biological processes and diseases. In cancer, it often acts as a tumor suppressor or is associated with prognosis. In rheumatoid arthritis, it contributes to inflammation. Studies on DERL3 help understand disease mechanisms and may provide potential therapeutic targets.
