Sting1, also known as STING or TMEM173, is an endoplasmic reticulum protein. It plays a fundamental role in the production of type I interferons (IFNs) and pro-inflammatory cytokines in response to microbial or host-derived DNA by activating multiple transcription factors. It is a key component of the cGAS-STING pathway, which is crucial for innate immunity, and is implicated in various biological processes such as autophagy and different cell death modalities [1].
In a study on crescentic glomerulonephritis, Sting1-/-mice with nephrotoxic nephritis had lower kidney cytokine expression, milder kidney infiltration by immune cells, and decreased disease severity. Pharmacological STING1 inhibition mirrored these findings, indicating that STING1 activation is a pathological mechanism in this disease [2].
In conclusion, Sting1 is essential for the cGAS-STING pathway in innate immunity, regulating autophagy and cell death. The use of Sting1 knockout mouse models has revealed its role in immune-mediated diseases like crescentic glomerulonephritis, providing insights into potential therapeutic targets for such conditions.
