Overview of Gene Research
Hemgn, also known as Hemogen in mouse, EDAG in human and RP59 in rat, is a gene encoding a hematopoietic-specific nuclear protein. It is involved in multiple biological processes. In hematopoiesis, it has been linked to the regulation of hematopoietic stem and progenitor cells (HSPCs) through negatively regulating the interferon-γ (IFN-γ) signaling pathway [1]. It also has potential roles in spermatogenesis as a testis-specific isoform exists [2].
In Hemgn-deficient mice, HSPCs show defective engraftment during bone marrow transplantation, with reduced homing, survival, and increased apoptosis. Transcriptome analysis of transplanted Hemgn -/- HSPCs reveals enrichment of gene sets related to IFN-γ signaling. Hemgn -/- HSPCs have enhanced responses to IFN-γ treatment and increased aging over time. Blocking IFN-γ signaling can rescue the engraftment defect of Hemgn -/- HSPCs [1].
In conclusion, Hemgn is a critical regulator for the successful engraftment and reconstitution of HSPCs in mice by negatively regulating IFN-γ signaling. The study of Hemgn knockout mouse models provides insights into its role in HSPC transplantation-related stress, which may help improve conditioning regimens and engraftment during HSPC transplantation [1].
