Overview of Gene Research
Hnf4aos, the mouse homolog of human HNF4A-AS1, is a long non-coding RNA. It is located between the P2 and P1 promoters of the HNF4A gene. Hnf4aos is involved in regulating the expression of genes, with potential importance in the development and function of digestive tissues [2].
In a mouse model, liver-specific knockdown of Hnf4aos led to increased serum aspartate and alanine transaminase levels, enlarged and degenerated hepatocytes, as well as increased endoplasmic reticulum stress markers 24 hours after ritonavir (RTV) administration. The aggravated liver injury was due to heightened Cyp3a11 expression, which is the mouse homolog of human CYP3A4. In vitro studies showed that HNF4A-AS1 (the human equivalent) mediated the interaction between heterogeneous nuclear ribonucleoprotein C and HNF4A, with the former promoting HNF4A degradation through the ubiquitination pathway, thus decreasing CYP3A4 expression and alleviating RTV-induced liver injury [1].
In conclusion, Hnf4aos plays a crucial role in regulating CYP3A4 expression, which is associated with RTV-induced hepatotoxicity. The liver-specific knockdown mouse model of Hnf4aos has revealed its important role in this disease-related process, providing insights into the underlying mechanism of RTV-induced liver injury [1].
