Overview of Gene Research
Pbx3, a homeodomain-containing transcription factor of the pre-B cell leukemia (PBX) family, has extensive roles in early development and some adult processes. It can form specific and stable DNA interactions without co-factors. PBX3 is involved in multiple signaling pathways, such as the P2X1-PBX3-BCAT1 pathway in leukemogenesis [1], and is related to metabolic reprogramming like promoting the pentose phosphate pathway through enhancing G6PD expression in colorectal cancer [2].
In a murine acute myeloid leukemia (AML) model, P2X1 deletion, which affects the P2X1-PBX3-BCAT1 pathway, impaired leukemia-initiating cell (LIC) self-renewal capacities and decreased functional LIC numbers without affecting normal hematopoiesis, revealing the role of PBX3 in leukemogenesis [1]. In colorectal cancer, PBX3 promotes cancer progression by enhancing G6PD expression, indicating its role in metabolic-based tumor development [2].
