Overview of Gene Research
Stim1, known as stromal interaction molecule 1, is a key component of the store-operated Ca2+ entry (SOCE) pathway. It is located on the endoplasmic reticulum membrane. Stim1 senses the Ca2+ content of the endoplasmic reticulum (ER) and plays a crucial role in the activation of Orai1 channels in the plasma membrane when ER Ca2+ stores are depleted. This process triggers Ca2+-dependent gene transcription, T-cell activation, and other essential cellular processes like gene regulation, cell growth, and death [1,3,4].
In hepatocellular carcinoma, Stim1 knockout-HCC cells generated by CRISPR-Cas9 showed that Stim1 down-regulation was associated with a shift from aerobic glycolysis towards AMPK-activated fatty acid oxidation (FAO), contributing to Snail1-driven metastasis and anoikis-resistance. Low Stim1 levels were associated with poor outcomes for HCC patients [2].
In conclusion, Stim1 is essential for regulating Ca2+ signaling through the SOCE pathway, which impacts a variety of cellular functions. The use of Stim1 knockout models in HCC has revealed its role in metabolic reprogramming during cancer metastasis, providing insights into potential therapeutic targets for this disease.
