Overview of Gene Research
Mrgprb2, a Mas-related G-protein-coupled receptor, is expressed on mast cells. It serves as a crucial mediator in various non-IgE-dependent mast cell activation pathways. Activation of Mrgprb2 can lead to mast cell degranulation, releasing substances like histamine, tryptase, and cytokines, which are involved in immune-related processes such as neuroimmune regulation, inflammation, and pain [2,5,7].
Mrgprb2-conditional knockout (Mrgprb2-/-) mice have been used in multiple studies to reveal its role. In an MC903-induced AD mouse model, Mrgprb2-/-mice showed milder phenotypes and less inflammatory infiltration, indicating that MRGPRB2-tryptase axis is involved in type 2 inflammation development in AD [1]. In Modic change models, Mrgprb2 knockout inhibited mast cell activation and reduced the degree of Modic changes, as Mrgprb2-activated mast cells regulate immune niches by influencing macrophage polarization [3]. In acute colitis models, loss of Mrgprb2-mediated signaling impaired DSS-induced neutrophil influx and impacted acute colitis progression [4]. In alcohol-withdrawal-associated headache models, Mrgprb2-deficient mice did not show headache-related behaviors during alcohol withdrawal, suggesting that Mrgprb2 in dura mater mast cells is involved in this process [6]. Baicalin-induced pseudo-allergy was absent in Mrgprb2-knockout mice, indicating baicalin induces pseudo-allergy via Mrgprb2 in mice [8].
In conclusion, Mrgprb2 plays a vital role in mast-cell-mediated immune responses, inflammation, and pain-related processes. Studies using Mrgprb2 KO/CKO mouse models have provided insights into its functions in diseases such as atopic dermatitis, Modic changes, acute colitis, alcohol-withdrawal-associated headache, and pseudo-allergic reactions, suggesting it could be a potential therapeutic target for these conditions.
