Overview of Gene Research
Nopchap1, also known as C12ORF45, is a PAQosome cofactor. The PAQosome, composed of the HSP90/R2TP chaperone and a prefoldin-like module, promotes the biogenesis of cellular machineries. Nopchap1 plays a crucial role in box C/D snoRNP biogenesis by acting as a bridge between NOP58 and PAQosome, and its study can be facilitated by genetic models [1].
In the research, Nopchap1 was found to directly interact with the CC-NOP domain of NOP58 and domain II of RUVBL1/2 AAA+ ATPases. Its interaction with RUVBL1/2 is disrupted upon ATP binding. In NOPCHAP1 KO cells, the expression of NOP58, but not NOP56 or PRPF31, is decreased, indicating that Nopchap1 is a client-loading PAQosome cofactor that selects NOP58 to promote box C/D snoRNP assembly [1]. Additionally, in a GWAS on Duroc × (Landrace × Yorkshire) pigs, Nopchap1 was identified as one of the 12 promising candidate genes associated with four bone weight traits, suggesting its potential role in pig bone weight-related genetic mechanisms [2].
In conclusion, Nopchap1 is essential for box C/D snoRNP assembly through its role as a PAQosome cofactor. The NOPCHAP1 KO cell experiments have revealed its specific function in this process. Moreover, its identification in pig GWAS implies a possible role in bone-related biological processes. These findings contribute to understanding its diverse biological functions and potential significance in related disease or trait-associated areas [1,2].
