Overview of Gene Research
ZDHHC18, a palmitoyltransferase, is involved in palmitoylation, an important post-translational modification. It participates in various biological processes and is associated with multiple pathways related to innate immunity, metabolism, and fibrosis. Genetic models, especially knockout (KO) mouse models, are valuable for studying its functions [1,2,3].
In innate immunity, Zdhhc18-deficient mice are resistant to DNA virus infection as ZDHHC18 negatively regulates cGAS-mediated innate immune responses by palmitoylating cGAS at C474, reducing its enzymatic activity and DNA binding ability [1]. In ovarian cancer, ZDHHC18-mediated palmitoylation of MDH2 at C138 activates mitochondrial respiration and accelerates cancer growth. In renal fibrosis, tubule-specific deletion of ZDHHC18 attenuates renal fibrosis as it normally promotes the process by regulating HRAS palmitoylation [2,3].
In conclusion, ZDHHC18 plays crucial roles in innate immunity, ovarian cancer malignancy, and renal fibrogenesis. The use of Zdhhc18 KO mouse models has provided significant insights into these disease-related biological functions, highlighting ZDHHC18 as a potential therapeutic target in related disease areas [1,2,3].
