Overview of Gene Research
Dbi, also known as diazepam binding inhibitor (DBI) or acyl-CoA binding protein (ACBP), is a phylogenetically conserved paracrine inhibitor of macroautophagy/autophagy, acting as a pro-aging molecule [1]. It can bind to the GABAA receptor and is involved in regulating lipid metabolism. Genetic loss-of-function models have been crucial in understanding its physiological roles in vivo [2].
In KO mouse models, knockout of ACB1 (yeast equivalent of human Dbi) induces autophagy and prolongs lifespan in an autophagy-dependent manner [1]. Antibody-mediated neutralization of Dbi in mice reduces signs of anthracycline-accelerated cardiac aging, such as the upregulation of the senescence marker CDKN2A/p16 and functional decline of the heart [1]. Also, in adipose tissue-specific Acbp/Dbi knockout mice, high-fat diet-induced weight gain is prevented [3].
In conclusion, Dbi plays essential roles in regulating autophagy, lipid metabolism, and body mass homeostasis. Its function has been revealed through gene knockout models, which have also demonstrated its significance in age-associated cardiovascular diseases and obesity-related conditions.
