Overview of Gene Research
HAMP, also known as hepcidin antimicrobial peptide, encodes a small peptide hormone that plays a vital role in regulating systemic iron homeostasis. Hepcidin binds to ferroportin, an iron exporter, leading to its internalization and degradation, thus controlling cellular iron efflux and completing an iron-regulating homeostatic loop [4,6]. This regulation is crucial for maintaining normal iron levels in the body, and disruptions can lead to various iron-related disorders.
In disease research, HAMP shows promise as a biomarker. In gastric cancer, high HAMP expression was associated with poor overall survival and was related to immune cell infiltration levels, suggesting it could serve as an independent prognostic biomarker through the immune pathway [1]. In non-small cell lung cancer, HAMP promoted cell proliferation, invasion, and migration by activating the NF-κB pathway, with non-coding RNAs regulating its activity [2]. In hepatocellular carcinoma, HAMP had a good ability to diagnose the disease, predict PD-1 treatment sensitivity, and was positively correlated with immune cell infiltration and immune checkpoint expression [3]. In addition, mutations in HAMP were found to cause an atypical form of juvenile hereditary hemochromatosis, which was misdiagnosed as type 1 diabetes initially [5].
