Overview of Gene Research
Pi16, also known as peptidase inhibitor 16, is a gene with diverse functions. Fibroblast-derived Pi16 is involved in immune-modulation, pain regulation, and cancer-related processes [1,2,3,4,5,6,7]. In pain regulation, it may act through influencing immune cell-fibroblast crosstalk; in cancer, it can have either tumor-suppressive or tumor-promoting roles depending on the context [1,3,4,5].

In gene knockout (KO) mouse models, Pi16-/-mice are protected against sustained inflammatory pain, likely due to reduced macrophage density and a bias towards anti-inflammatory CD206hi macrophages in the pain neuroaxis [1]. In neuropathic pain, Pi16-/-mice show reduced endothelial barrier permeability, lower leukocyte infiltration, and are protected against neuropathic pain, suggesting Pi16 promotes pain by mediating fibroblast-endothelial barrier cross-talk [7]. Conditional knockout (CKO) of Pi16 in regulatory T cells (Tregs) enhances their differentiation and suppressive functions, protecting mice from autoimmune arthritis and dextran sulfate sodium-induced colitis [8].

In conclusion, Pi16 plays crucial roles in pain, immune regulation, and cancer. KO and CKO mouse models have revealed its functions in inflammatory and neuropathic pain, as well as autoimmune diseases, providing insights into potential therapeutic targets for chronic pain, autoimmune, and inflammatory diseases.
