Overview of Gene Research
Gusb, encoding β -glucuronidase (GUS), is essential for degrading glycosaminoglycans (GAGs), including heparan sulfate, dermatan sulfate, and chondroitin-4,6-sulfate. Deficiency in GUSB leads to mucopolysaccharidosis VII (MPS VII; Sly syndrome), an autosomal recessive disorder associated with various symptoms such as mental retardation, short stature, and hepatosplenomegaly [3].
In an endometriosis C57BL6 mouse model, gut dysbiosis-derived GUSB promoted the development of endometriosis by enhancing the proliferation and invasion of endometrial stromal cells and causing macrophage dysfunction [1]. In hepatocellular carcinoma, over-expression of GUSB led to primary resistance of anti-PD1 therapy as it facilitated cell proliferation, invasion, and migration and downregulated PD-L1 expression by promoting miR-513a-5p [2].
In conclusion, Gusb is crucial for GAG degradation, and its dysfunction is associated with MPS VII. Studies in mouse models have also revealed its role in endometriosis and hepatocellular carcinoma, highlighting its significance in understanding these disease mechanisms.
