Overview of Gene Research
Rbm47, an RNA-binding protein, is crucial for embryonic endoderm development [1]. It contains three RNA recognition motif (RRM) domains and is conserved in vertebrates. Rbm47 is involved in post-transcriptional regulation of gene expression, immune homeostasis, and various signaling pathways such as Wnt/β-catenin [3].
Intestine-specific Rbm47-knockout (Rbm47-IKO) mice studies show that Rbm47 regulates intestinal injury, proliferation, and tumorigenesis. Rbm47-IKO mice exhibit increased proliferation, abnormal villus morphology, and adapt better to radiation injury and chemical-induced colitis. They are also protected against colitis-associated cancer, but aged Rbm47-IKO mice develop spontaneous polyposis [1]. In lung, K329R knockin (KI) mice with defective RBM47-ISGylation display heightened susceptibility to LPS-induced acute lung injury and lung tumorigenesis, indicating Rbm47-ISGylation's role in maintaining lung homeostasis [2].
In conclusion, Rbm47 plays essential roles in embryonic development, immune homeostasis, and tissue-specific functions like in the intestine and lung. Gene-knockout mouse models, such as Rbm47-IKO and K329R KI mice, have been instrumental in revealing its role in intestinal injury, tumorigenesis, and lung-related diseases, providing insights into its potential as a therapeutic target in these disease areas.
