Overview of Gene Research
Pm20d1, or Peptidase M20 Domain Containing 1, is a secreted enzyme that catalyzes the condensation of fatty acids and amino acids into bioactive lipids called N-acyl amino acids (NAAA) [3]. It is involved in multiple biological processes, with its role in metabolism and neurodegenerative diseases being of particular interest. PM20D1 is also associated with pathways like TRPV4-mediated calcium influx regulation [1]. Genetic models are valuable for studying its functions.
In a mouse model of synucleinopathy, overexpression of PM20D1 alleviated α-Syn pathology, dopaminergic neurodegeneration, and motor impairments, suggesting its protective role against Parkinson's disease progression [1]. In Alzheimer's disease, genetically increasing PM20D1 levels decreased AD-related pathologies, while decreasing its levels accelerated them, reinforcing its likely neuroprotective role [2,4]. In polycystic ovary syndrome-insulin resistance (PCOS-IR), loss of adipocyte PM20D1 aggravated insulin resistance (IR), and quercitrin, by upregulating PM20D1, corrected reproductive and metabolic abnormalities [5].
In conclusion, PM20D1 plays essential roles in metabolism-related processes and neurodegenerative diseases. Model-based research, especially through overexpression and gene-altering studies in mouse models, has revealed its protective functions in Parkinson's and Alzheimer's diseases, as well as its importance in PCOS-IR. These findings contribute to understanding the biological mechanisms of these diseases and may offer potential therapeutic targets.
