Overview of Gene Research
AREL1, an antiapoptotic E3 ubiquitin ligase, belongs to the "Other" subfamily of HECT E3 ligases. It plays a crucial role in regulating apoptosis, necroptosis, and inflammation by ubiquitinating and degrading various proteins. It is involved in pathways such as those related to TGF-β -induced apoptosis, TNF -induced necroptosis, and IL -1β turnover [1,2,3].
In vascular endothelial cells, AREL1 is downregulated by miR-320b, and its overexpression inhibits TGF-β-induced apoptosis by downregulating SMAC, a proapoptotic protein [1]. In TNF-induced necroptosis, AREL1 ubiquitinates and promotes the degradation of MTX2, thus inhibiting necroptosis. Inhibition of AREL1-dependent ubiquitination of MTX2 could potentially sensitize tumor cells to TNF-induced necroptosis [2]. Additionally, AREL1, along with UBE2L3 and TRIP12, limits inflammation by promoting pro-IL-1β turnover [3].
In conclusion, AREL1 is essential in regulating apoptosis, necroptosis, and inflammation. Studies on its function in various disease-related processes, such as arteriosclerosis and tumor cell necroptosis, provide potential therapeutic targets for these diseases.
