Overview of Gene Research
Safb, also known as scaffold attachment factor B, is a conserved RNA-binding protein essential for early mammalian development. It is involved in multiple biological pathways, such as gene regulation, DNA repair, and RNA processing. It has been implicated in various biological processes including cell fate determination, apoptosis, and maintaining genome integrity [1,2,4]. Genetic models like KO/CKO mouse models are valuable for studying its functions.
In mouse embryonic stem cells, knockout of Safb/Safb2 led to differential expression of around 1000 genes related to apoptosis, cell division, and cell migration, indicating its role in promoting the expression of certain genes by binding nascent RNA [1].
In adult hippocampal neural stem cells, conditional deletion of Safb increased the expression of NFIB and oligodendrocyte formation, showing that Safb blocks oligodendrocyte production by enhancing Drosha-dependent cleavage of Nfib mRNA [2].
In acute myeloid leukemia (AML), perturbation of SAFB phenocopied HOXA9 knockout, decreasing AML proliferation and increasing differentiation and apoptosis, suggesting that SAFB forms a repressive complex with HOXA9 to maintain AML [3].
