Overview of Gene Research
Crip3 is a member of the cysteine-rich intestinal protein (CRIP) family, which belongs to the highly conserved Lin-1, Isl1, Mec3/double zinc finger protein family [2]. The CRIP family plays important roles in cellular epithelial-mesenchymal transition, cell death, and tumor progression, and participates in multiple signaling pathways [2].
Analysis of RNA-seq data under oxidative stress conditions revealed that Crip3 was particularly downregulated, suggesting a relationship between oxidative stress and zinc homeostasis [3]. In a study on age-related hearing loss, an association was detected between a common variant in Crip3 and age-related hearing loss [1].
In prostate cancer research, a prognostic multigene methylation panel including CRIP3 in radical prostatectomy specimens was developed, and this panel could prognosticate late biochemical recurrence and the need for therapy after surgery [4]. Also, a 4-gene methylation classifier panel (including CRIP3) in urinary sediment DNA was able to predict disease progression in prostate cancer patients on active surveillance [9]. In addition, a 3-marker panel consisting of miR-24, miR-30c and CRIP3 methylation was significant for predicting reclassification of prostate cancer patients on active surveillance [8].
In the study of heart failure, CRIP3 was identified as a novel contributor to heart failure pathogenesis through combined histological and clinical analysis [5].
In squamous cell carcinoma of the head and neck, an 8-gene HPV-related prognostic model including CRIP3 was established, which might serve as a candidate biomarker and therapeutic target [6]. A genetic association study found an association between a variant near CRIP3 and pulse pressure [7].
In conclusion, Crip3 is involved in multiple biological processes and diseases. Its downregulation under oxidative stress implies its role in maintaining zinc homeostasis. In diseases such as age-related hearing loss, prostate cancer, heart failure, and squamous cell carcinoma of the head and neck, Crip3 shows potential as a biomarker or a factor in disease pathogenesis. The studies on Crip3 in these disease areas contribute to understanding the underlying molecular mechanisms and provide potential targets for diagnosis and treatment.
