Overview of Gene Research
NAPG, encoding gamma-SNAP (Soluble NSF Attachment Protein gamma), is located on chromosome 18p11. It is one of three SNAPs and plays a role in cellular processes required for neurotransmission in the central nervous system [5].
In a study of a large Chinese pedigree with hereditary hemorrhagic telangiectasia (HHT), a mutation (c.784A > G) in NAPG was identified. Functional prediction suggested this mutation was deleterious and might change the conformational stability of the NAPG protein, potentially leading to HHT [1]. In the Chinese Han population, several SNPs in NAPG were associated with bipolar disorder. Rs473938 and rs2290279 showed significant association in allele and genotype frequencies, and some haplotypes were also associated with bipolar disorder [2]. Another study analyzing SNPs in NAPG also found nominal, statistically significant associations with bipolar disorder at the genotype frequency level for three SNPs [5].
In fetal growth restriction (FGR), NAPG was among the top 5 altered mRNA genes, with its up-regulation detected in newborn umbilical cord blood, maternal serum, and placental tissue [3]. Mendelian randomization analysis indicated ZG16B, PEX14 in blood and ZG16B, NAPG in prostate tissue were potential drug targets for prostate cancer, and a phenome-wide association study showed possible side effects of NAPG as a drug target [4].
In conclusion, NAPG is involved in neurotransmission-related cellular processes. Studies have revealed its potential associations with diseases such as HHT, bipolar disorder, FGR, and prostate cancer, highlighting its importance in understanding the genetic basis of these diseases and potentially developing related treatments.
