Overview of Gene Research
Wnt3a, a member of the Wnt family of secreted glycoproteins, is crucial in regulating multiple cellular functions such as self-renewal, proliferation, differentiation, and motility. It exerts its effects mainly through the canonical Wnt/β-catenin signaling pathway, which is involved in numerous biological processes including development, tissue homeostasis, and disease [3].

In lung injury, Wnt3a-loaded extracellular vesicles (EVs) can promote alveolar epithelial regeneration. The engineered Wnt3aWG EVs activate Wnt signaling, enhance cell growth after human alveolar epithelial cell injury, and restore impaired pulmonary function in an elastase-induced emphysema model [1]. In colorectal cancer, Wnt3a is associated with angiogenesis and cancer progression, making it a promising target for treatment [2]. Regarding cartilage repair, WNT3a-loaded exosomes can penetrate cartilage, activate canonical WNT signaling, and improve the repair of osteochondral defects in mice, while recombinant WNT3a fails to activate the signaling in cartilage [4]. In the context of osteoarthritis, knockdown of Wnt3a in rat chondrocytes promotes collagen type II expression, suggesting it could be a novel therapy method [5]. In cardiac hypertrophy, TGFβ1 can upregulate Wnt3a expression in a Smad-dependent manner, and there is crosstalk between TGFβ1 and the canonical Wnt/β-catenin pathways [6]. Also, in hepatocarcinogenesis, the overexpression of Wnt3a in a rat model was associated with the process, and it could be a promising biomarker for early-stage HCC monitoring [7].

In conclusion, Wnt3a plays diverse and significant roles in various biological processes and disease conditions. Studies using different animal models, such as the mouse models in lung injury, cartilage repair, and cardiac hypertrophy, have revealed its functions in tissue regeneration, disease progression, and potential as a biomarker or therapeutic target. These findings contribute to a better understanding of the underlying mechanisms of related diseases and provide new perspectives for treatment strategies.
