CRISPR/Cas and TurboKnockout: Technologies for Rapid Generation of Knockout/Knockin Mouse Models

While genetically engineered mouse/rat models have been used widely in biomedical research, the design and generation of these models can be complicated, time-consuming and costly. In this presentation, we will briefly review the traditional pronuclear injection based transgenics and ES cell homologous recombination based gene targeting, and go over the recently developed CRISPR-mediated genome editing technology, with an emphasis on the pros and cons of each technology and their applications. Then will introduce TurboKnockout, the newest ES-cell based gene targeting technology which allows the generation of conditional knockout/knockin mouse models in as fast as 6 months, 4-6 months shorter than the standard approach. Lastly, we will highlight a novel online tool called VectorBuilder, which promises to revolutionize the way DNA vector cloning is done.
