Strain Description
Tumor-associated calcium signal transducer 2 (TROP2), encoded by the TACSTD2 gene, is an important type I transmembrane glycoprotein that plays a critical regulatory role in multiple cellular biological processes. As a typical cell surface receptor, TROP2 induces intracellular calcium release by transducing extracellular signals, thereby regulating key biological behaviors such as cell proliferation, adhesion, migration, and differentiation [1]. Under normal physiological conditions, TROP2 is highly expressed during embryonic development (particularly in fetal tissues and trophoblast cells), while only basal expression levels are maintained in some adult normal epithelial tissues. However, TROP2 expression is significantly and aberrantly upregulated in a variety of human malignant tumors, a feature that makes it an important tumor marker [2]. Studies have shown that dysregulated TROP2 expression can promote tumor initiation, progression, and metastasis through the activation of multiple signaling pathways [3]. In addition to its oncogenic role, germline mutations in the TACSTD2 gene have also been confirmed as the causative factor for gelatinous drop-like corneal dystrophy, a rare autosomal recessive disorder [4]. Notably, TROP2 expression is restricted in most normal adult tissues but is highly enriched in various solid tumors. This highly selective expression pattern makes it a highly promising therapeutic target [3].
The huTROP2(TACSTD2)(2) mouse is a humanized mouse model generated through a gene editing strategy, in which the exon 1 plus flanking region of mouse Tacstd2 gene was replaced with the exon 1 plus flanking region of human TACSTD2 gene. The humanized sequence contains the UTR region of the human TACSTD2 gene. This strain can be used for studying the pathogenesis of various solid tumors, as well as for the screening, development, and in vivo evaluation of TACSTD2-targeted drugs.
A comparable hTROP2(TACSTD2) mouse (signal peptide and extracellular domain-humanized model; Catalog No.: C001718) has also been developed by Cyagen. The two strains differ in the composition of the humanized expression elements: only the signal peptide and extracellular domain are humanized in C001718, whereas the human TACSTD2 exon 1 (including the UTR) together with its upstream and downstream flanking sequences is humanized in this model. The appropriate model should be selected according to the research purpose.
