Strain Description
NKG mice (Catalog No.: C001316) are a type of severely immunodeficient mouse independently developed by Cyagen through knockout of the Il2rg gene on the NOD-Scid background. This strain lacks mature T, B, and NK cells, has reduced complement activity, and its macrophages exhibit weak phagocytosis of human cells. Therefore, NKG mice can efficiently engraft human hematopoietic stem cells (HSC), peripheral blood mononuclear cells (PBMC), patient-derived tumor xenografts (PDX), or adult stem cells and tissues.
A series of molecules encoded by the mouse major histocompatibility complex (MHC) play a central role in antigen processing and T cell-mediated immune responses. These molecules are roughly divided into two classes: class I molecules (e.g., H2-K1 and H2-D1), which are widely expressed on the surface of all nucleated cells and primarily function to present endogenous antigens to CD8+ cytotoxic T cells; and class II molecules (e.g., H2-Ab1), which are mainly expressed by professional antigen-presenting cells (APCs), including macrophages, B cells, and dendritic cells, and are responsible for presenting exogenous antigens to CD4+ helper T cells [1-2].
Previous studies have shown that double knockout (dKO) of MHC class I and class II molecules in mice can block the xenogeneic antigen presentation process, thereby alleviating graft-versus-host disease (GvHD) and enabling long-term engraftment of human immune cells [3-5]. NKG-MHC-dKO(H2-K1/H2-Ab1/H2-D1) mice (Catalog No.: C001705), generated by targeted deletion of the key MHC class I molecules (H2-K1 and H2-D1) and the class II molecule (H2-Ab1), can circumvent the immune mismatch that triggers GvHD.
The huPBMC-NKG-MHC-dKO mouse is an immune system-humanized model constructed by transplanting human PBMC into NKG-MHC-dKO(H2-K1/H2-Ab1/H2-D1) mice (Catalog No.: C001705). This model may help reduce GvHD associated with immune mismatch after PBMC transplantation and is suitable for long-term in vivo research and evaluation of human immune system reconstitution.

Application Area
For in vivo studies that require mature T cells in areas such as tumor immunity, GvHD, hematological diseases, and infectious diseases (e.g., HIV/AIDS), thereby facilitating the development of new drugs related to immunotherapy.
